Phage DNA Kit
Product Overview:
The Phage DNA Isolation Kit provides a fast and efficient method for isolating total DNA from bacteriophages propagated in liquid bacterial cultures. The procedure avoids the use of phenol, chloroform, or cesium chloride, providing a safe and convenient workflow.
The spin-column based procedure can be completed in under 45 minutes and is optimized for small volumes of phage supernatant (1 mL). The resulting DNA is of high integrity and is compatible with a variety of downstream applications, including Southern blotting, restriction fragment length polymorphism (RFLP), sequencing, cloning, and real-time PCR.
Purification Technology:
The kit uses spin column chromatography, which selectively binds DNA while removing proteins and other contaminants. The workflow includes:
- Starting with clarified phage supernatant free of bacterial debris
- Lysis of phage particles using heat and chemical treatment
- Addition of isopropanol and loading of the lysate onto a spin column
- Selective DNA binding under ionic conditions
- Washing to remove RNA, proteins, and other impurities
- Elution of purified phage DNA ready for downstream use
