This Fungi/Yeast Genomic DNA Isolation Kit is designed for rapid preparation of genomic DNA from viable yeast cells, fungal spores or mycelium, and Gram-positive bacteria. Genomic DNA is efficiently extracted using a combination of heat treatment, detergents, and bead-based homogenization. The purified DNA is fully digestible with tested restriction enzymes and is compatible with downstream applications such as PCR and Southern blot analysis. DNA yields may vary depending on cell density and species. An optional lyticase treatment is included to improve yields for certain fungal and yeast species. Preparation time for a single sample is under 30 minutes, and the kit provides sufficient materials for 50 preparations.

Purification Technology

Purification is based on spin column chromatography. The process begins by adding lysis buffer to the sample, with an optional RNase treatment if RNA-free genomic DNA is required. The sample is then transferred to a bead tube and vortexed for 5 minutes to efficiently homogenize the cells and extract the DNA. After centrifugation, the supernatant is collected in a DNase-free tube. Ethanol and a binding solution are added, and the mixture is applied to a spin column. The column selectively binds nucleic acids based on ionic concentrations, allowing DNA to bind while proteins are removed in the flowthrough or retained on the resin. Bound DNA is washed twice with the provided wash solution and eluted using the elution buffer.

The purified genomic DNA is free of inhibitors and suitable for sensitive downstream applications, including PCR and Southern blotting.
